The Novartis Institutes for BioMedical Research (NIBR) is the innovation engine of Novartis. We collaborate across scientific and organizational boundaries, with a focus on powerful new technologies that have the potential to help produce therapeutic breakthroughs for patients.
The translational research group in the Autoimmunity, Transplantation and Inflammation Disease Area utilizes state-of-the-art technology and computational biology to improve our understanding of underlying, causative pathways in autoimmune and inflammatory diseases. We have particular interest in the role of CD40-CD154 interactions in disease, and we have generated and characterized a pathway blocking, non-depleting anti-CD40 antibody (CFZ533) currently in clinical development for allograft transplantation and autoimmune disease. We also focus on the biochemical, molecular and cellular outcomes of CD40-CD154 interactions, and data from a recent in-house, genome-wide CRISPR screen point to pathways involved in regulation of receptor internalization and recycling as key regulators of signaling downstream of CD40. We therefore want to recruit a postdoctoral fellow to systematically examine the relationship between CD40 receptor internalization and multiple downstream signaling pathways in different CD40 expressing cell types, including following up on various novel hits from the previously mentioned CRISPR screen. If a link between receptor-ligand internalization could be established for CD40 and other TNF-R family members, it would represent a significant advance in our understanding of pathway signaling downstream of this family of receptors. To enable these experiments, we are equipped with state-of-the-art flow cytometry, imaging, biochemical and molecular technologies as well as expertise in CRISPR, immunology and translational research, as well as a variety of tools to interrogate CD40 expression, and pathway activation.
The successful candidate should have experience in the cellular and biochemical regulation of endocytosis and endosomal trafficking as well as knowledge of immunology. Expertise in imaging and cutting-edge microscopy will be highly regarded. They will join a vibrant scientific community and collaborate with other postdoctoral fellows mentored by a range of scientists across disciplines and platforms, including chemistry, structural biology, molecular biology, functional genomics, neurophysiology, computational biology and translational medicine. Postdoctoral scholars at NIBR have a unique opportunity to conduct innovative, interdisciplinary research and are expected to publish their results in leading journals.
